方案概述
前言:
食品脂肪酸组成检测是评价食品营养价值与品质安全的核心环节。通过对食品中饱和脂肪酸、单不饱和脂肪酸以及多不饱和脂肪酸的精确分析,可以有效评估食品的膳食结构比例,为营养标签标识提供科学依据。
本篇文章参考了《GB 5009.168-2016 食品安全国家标准 食品中脂肪酸的测定》,使用英国365集团官网GC6100气相色谱配FID检测器,对芝麻油中的脂肪酸进行测试。

2.实验试剂及辅助设备
2.1 实验试剂
氢氧化钠(分析纯);
甲醇(色谱纯);
正庚烷(色谱纯);
15%三氟化硼甲醇溶液;
无水硫酸钠(分析纯);
37种脂肪酸甲酯标样。
2.2 前处理设备
分析天平(万分之一);
水浴锅;
旋转蒸发仪;
实验室常用玻璃器皿等。
3.实验方法
3.1 气相色谱仪参考条件
柱温:初始温度50℃,保持1min,以30℃/min升温至200℃,保持3.5min;
分流比:50:1;
进样口温度:260℃;
检测器温度:250℃;
空气流量:300mL/min;
氢气流量:40mL/min;
进样体积:1μL。
3.2 样品处理
称取适量芝麻油样品(0.1g-0.25g)于100mL烧瓶中;加入2%氢氧化钠甲醇溶液8mL,连接回流冷凝管,80℃水浴回流,直至油滴消失;7mL15%三氟化硼甲醇溶液从冷凝管上端加入,用少量水润洗,回流2min左右;停止加热,迅速冷却至室温;吸取上层正庚烷提取溶液5ml,至25ml试管中,加入3-5g无水硫酸钠,振摇1min,离心5min(或静置5min),吸取上层清液待测定。

说明:从图2和图3可看出37种脂肪酸在DB-FastFAME(30m*0.25mm*0.25)上出峰时间比CP-Sil 88(100m*0.25mm*0.2um)快,且分离效果良好,下文中重复性和实际样品测试结果均为用DB-FastFAME(30m*0.25mm*0.25)测试所得。
4.2 标准样品重复性测试
用标准品重复进样5次,得到5针标准样品重复性。

从表中数据可知各组分面积RSD均小于0.5%,时间重复性RSD均小于0.015%。
5.结论
本次测试采用英国365集团官网气相色谱仪GC6100配置FID检测器分别对食品中脂肪酸进行检测。实验结果显示,37种脂肪酸甲酯能很好分离,做标样重复性,37种组分面积RSD均小于1.1%,满足实验分析需求。芝麻油经甲酯化后测试,用从结果可知,芝麻油的主要脂肪酸包括油酸(38.8%)、亚油酸(44.1%)、棕榈酸(9.6%)和硬脂酸(5.6%)。 其中,油酸和亚油酸作为不饱和脂肪酸占比超过80%,是芝麻油的核心成分,赋予其抗氧化性和营养价值;棕榈酸和硬脂酸为饱和脂肪酸,占比相对较低。对样品进行重复性测试,各组分面积RSD均小于0.5%,时间重复性RSD均小于0.015%。
从实验数据可知,英国365集团官网GC6100测试脂肪酸时,分离度、时间重复性,面积重复性均可满足《GB 5009.168-2016 食品安全国家标准 食品中脂肪酸的测定》需求。
